The mechanism of action of antileishmanial compounds is poorly understood. Ultrastructural changes in Leishmania tropica within human macrophages exposed in vitro to Pentostam, pentamidine, amphotericin B, WR 6026, ketoconazole, and Formycin B were examined in these experiments. In Pentostam-treated cultures, some organisms exhibited diminished definition of mitochondrial and other membranes, while other organisms had completely disintegrated. Pentostam-exposed macrophages demonstrated loss of membrane definition in the absence of further alterations; it is therefore hypothesized that impaired macrophage membrane function may contribute towards the effect of this drug against macrophage-contained organisms. Leishmania parasites in pentamidine-treated cultures initially demonstrated swollen kinetoplasts and fragmentation of the kinetoplast DNA core. The initial observed effect of the other four drugs on the parasites was cytoplasmic condensation. These ultrastructural studies suggest that all five non-antimonial drugs may have different mechanisms of action than antimony (Pentostam) against Leishmania.  相似文献

A detailed study was made of the content per leaf lamina ofseveral biochemical components during growth and the entirelifespan of the primary leaf of bean, Phaseolus vulgaris L.,var. Dwarf Horticultural Bush. Total DNA measured spectrofluorometricallywas retained even after abscission and withering of the lamina.Possible reasons for the lack of DNA degradation in very oldleaves are presented. Using improved extraction methods it was found that both putativecytoplasmic polyribosomes and the total organelle plus cytoplasmicribosomal RNAs rose to a peak during leaf growth, declined rapidly,and finally levelled off during maturity and senescence. Even37 d old senescent leaves retained polysomes as large as hexamers.Organelle rRNA peaked late in the growth phase and about 4 dafter the peak in cytoplasmic rRNA. During leaf maturity andsenescence, both of these rRNA types decreased in parallel.Total protein and chlorophyll peaked at different times duringleaf growth, declined at a steady rate during maturity, andthen declined faster in the senescence phase. 1. Human activities affect fish assemblages in a variety of ways. Large‐scale and long‐term disturbances such as in‐stream dredging and mining alter habitat and hydrodynamic characteristics within rivers which can, in turn, alter fish distribution. Habitat heterogeneity is decreased as the natural riffle–pool–run sequences are lost to continuous pools and, as a consequence, lotic species are displaced by lentic species, while generalist and invasive species displace native habitat specialists. Sediment and organic detritus accumulate in deep, dredged reaches and behind dams, disrupting nutrient flow and destroying critical habitat for habitat specialist species. 2. We used standard ecological metrics such as species richness and diversity, as well as stable isotope analysis of δ13C and δ15N, to quantify the differences in fish assemblages sampled by benthic trawls among dredged and undredged sites in the Allegheny River, Pennsylvania, U.S.A. 3. Using mixed‐effects models, we found that total catch, species richness and diversity were negatively correlated with depth (P < 0.05), while species richness, diversity and proportion of species in lithophilic (‘rock‐loving’) reproductive guilds were lower at dredged than at undredged sites (P < 0.05). 4. Principal components analysis and manova revealed that taxa such as darters in brood hider and substratum chooser reproductive guilds were predominantly associated with undredged sites along principal component axis 1 (PC1 and manova P < 0.05), while nest spawners such as catfish and open substratum spawners including suckers were more associated with dredged sites along PC2 (P < 0.05). 5. Stable isotope analysis of δ13C and δ15N revealed shifts from reliance on shallow water and benthic‐derived nutrients at undredged sites to reliance on phytoplankton and terrestrial detritus at deep‐water dredged sites. Relative trophic positions were also lower at dredged sites for many species; loss of benthic nutrient pathways associated with depth and dredging history is hypothesised. 6. The combination of ecological metrics and stable isotope analysis thus shows how anthropogenic habitat loss caused by gravel dredging can decrease benthic fish abundance and diversity, and that species in substratum‐specific reproductive guilds are at particular risk. The effects of dredging also manifest by altering resource use and nutrient pathways within food webs. Management and conservation decisions should therefore consider the protection of relatively shallow areas with suitable substratum for spawning for the protection of native fishes.  相似文献

The evolutionary effects of Southern Hemisphere Pleistocene oceanographic conditions — marked by fluctuations in sea levels and water temperatures, and redirected currents — are poorly understood. The southeastern tip of Australia presents an intriguing model system for studying the biological impacts of palaeoceanography. In particular, contrasting oceanographic conditions that existed on eastern vs. western sides of the Bassian Isthmus during Pleistocene glacial periods allow for natural comparisons between putative refugial vs. re-invading populations. Whereas many western Tasmanian marine taxa were likely eliminated by cold subantarctic water during the last glacial period, eastern Tasmanian populations would have persisted in relatively warm temperatures mediated by the ongoing influence of the East Australian Current (EAC). Here we test for the effects of contrasting palaeoceanographic conditions on endemic bull kelp, Durvillaea potatorum , using DNA sequence analysis (COI; rbc L) of more than 100 individuals from 14 localities in southeastern Australia. Phylogenetic reconstructions reveal a deep (maximum divergence 4.7%) genetic split within D. potatorum , corresponding to the 'eastern' and 'western' geographical regions delimited by the Bassian Isthmus, a vicariant barrier during low Pleistocene sea levels. Concordant with the western region's cold glacial conditions, samples from western Tasmania and western Victoria are genetically monomorphic, suggesting postglacial expansion from a mainland refugium. Eastern samples, in contrast, comprise distinct regional haplogroups, suggesting the species persisted in eastern Tasmania throughout recent glacial periods. The deep east–west divergence seems consistent with earlier reports of morphological differences between 'western' and 'eastern' D. potatorum , and it seems likely that these forms represent reproductively isolated species.  相似文献

1. Shallow lakes are often cited as classic examples of systems that exhibit trophic cascades but, whilst they provide good model systems with which to test general ecological theory and to assess long‐term community change, their food web linkages have rarely been resolved, so changes associated with the structure and dynamics of the ecological network as a whole are still poorly understood. 2. We sought to redress this, and to demonstrate the potential benefits of integrating palaeolimnological and contemporary data, by constructing highly resolved food webs and stable isotope derived measures of trophic interactions and niche space, for the extant communities of two shallow U.K. lakes from different positions along a gradient of eutrophication. The contemporary surface sediment cladoceran and submerged macrophyte assemblages in the less enriched site, Selbrigg Pond, matched the palaeolimnological assemblages of the more enriched site, Felbrigg Hall Lake, in its more pristine state during the 1920s. Thus, Selbrigg was a temporal analogue for Felbrigg, from which the consequences of long‐term eutrophication on food web structure could be inferred. These data represent the first steps towards reconstructing not only past assemblages (i.e. nodes within a food web), but also past interactions (i.e. links within a food web): a significant departure from much of the previous research in palaeolimnology. 3. The more eutrophic food web had far fewer nodes and links, and thus a less reticulate network, than was the case for the more pristine system. In isotopic terms, there was vertical compression in δ15N range (NR) and subsequent increased overlap in isotopic niche space, indicating increased trophic redundancy within Felbrigg. This structural change, which was associated with a greater channelling of energy through a smaller number of nodes as alternative feeding pathways disappear, could lead to reduced dynamic stability, pushing the network towards further simplification. These changes reflected a general shift from a benthic‐dominated towards a more pelagic system, as the plant‐associated subweb eroded. 4. Although these data are among the first of their kind, the palaeo‐analogue approach used here demonstrates the huge potential for applying food web theory to understand how and why these ecological networks change during eutrophication. Furthermore, because of the rich biological record preserved in their sediments, shallow lakes represent potentially important models for examining long‐term intergenerational dynamics, thereby providing a means by which models and data can be integrated on meaningful timescales – a goal that has long proved elusive in food web ecology.  相似文献

Several complexes of species in Sphagnum (peat mosses) originated through hybridization and allopolyploidy, suggesting that these processes have played a major evolutionary role in this genus. The Sphagnum subsecundum complex includes gametophytically haploid and diploid species in North America. Analyses of 12 microsatellite loci and sequences from two plastid DNA markers show that the evolutionary history of this group is substantially more complex than previously thought. Two taxonomic species, Sphagnum lescurii and Sphagnum inundatum, include both haploid and diploid populations. Within each ploidal level, S. lescurii and S. inundatum are not genetically differentiated. The diploid taxa show patterns of fixed heterozygosity for the microsatellite markers, consistent with an allopolyploid origin. Diploid S. lescurii is an allopolyploid between haploid S. lescurii and (haploid) S. subsecundum. Sphagnum carolinianum is an allopolyploid between haploid S. lescurii and an unknown parent. We detected homoploid hybridization between the haploids Sphagnum contortum and S. subsecundum. Finally, we report three samples of diploid Sphagnum platyphyllum (otherwise haploid) that have an allopolyploid origin involving north‐eastern haploid S. platyphyllum and an unidentified taxon. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 135–151.  相似文献

The taxonomic and evolutionary affinities of Southern hemisphere smooth‐shelled blue mussels are unclear, with studies using different marker types having identified different relationships among various geographic regions. Using an existing and a new molecular assay, the present study builds on previous work to test the distribution of blue mussels native to and introduced to the Southern hemisphere. Populations of Mytilus were sampled from New Zealand, Australia, and Chile. The nuclear‐DNA marker Me 15/16 was used to identify the taxonomic status of 484 individuals. A new restriction fragment length polymorphism (RFLP) assay was used to identify the hemisphere of origin for a subset of Mytilus galloprovincialis. The Me15/16 marker identified 478 pure M. galloprovincialis from Southern hemisphere sites and six Mytilus edulis/M. galloprovincialis hybrids from the Auckland Islands (New Zealand) and Chile. A cytoplasmic RFLP identified Northern hemisphere M. galloprovincialis in almost every Southern hemisphere region. The presence of native M. galloprovincialis at high latitudes (up to 52°S) has implications for our understanding of environmentally induced selective constraints considered to determine species distributions. Widespread occurrence of invasive Northern hemisphere blue mussels in the Southern hemisphere is documented for the first time. Identification of inter‐specific hybrids (M. edulis × M. galloprovincialis) in Chile and in the Auckland Islands (subantarctic New Zealand) illustrates that environments ranging from international ports to remote protected locations are vulnerable to bioinvasion. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 101 , 898–909.  相似文献
